My interest in NixOS on the RPI4 is in an automated build that incorporates the latest upstream open source graphics stack for this board, and specifically the V3D kernel driver. Is that already incorporated into this configuration?

I don't think so, since I make this config pinned to NixOS of 2021.
But there is a possibility to have the kernel customized, however I think you need to build it yourself if this doesn't exists in public nix cache.
I have no way to test this myself since RPI4 is (ironically) insanely expensive here.
